I hope you don't mind some writing advice. You really don't have to give details about characters like what kind of pony they are and their colors. Readers already know those things, or at least they should, and I doubt that many don't. If it's an alternate universe thing and their class and appearance has been changed, then you could give that detail. Since this is anthro, giving body descriptions is fine.

This is interesting for sure. I think the biggest thing is Spike should be against sleeping with her at the start. I mean very against it. What dude would break the guy code by sleeping with a friend's mom when you were send there to help them. That put you on grounds of getting your ass beat half to death for sure even if said mom came on to you first. Guy code is law you break it you are trash lol

Hey there is a reason they are called glass cannons lol. He doesn't have to be physically stronger if his magic can make up for it. And it seem pretty clear this isn't warrior I have fought hundreds of battles Spike for sure. And he is just as into magic as Twilight I would find it hard he believe he hasn't studied how to take down a dragon when he lives somewhere that would be considered a dragons wet dream lol. But I am sure you will find a way to make it so Spike isn't the asshole in this.

Rarity is too easy...everyone's on the tick of that Spike lost his virginity to his long-standing crush. I would not be surprised if that were the case though.
Personally my choices would be --and this is a surprise for me--Pinkie and then (not that surprising) Fluttershy. What can I say, I do still love Sparity, but I really like Flutters and Spike together a bit as well. Why can't we ever see more stories with them together, or even one episode?? Just like Starlight and Spike work well together, I think that Fluttershy and Spike would surprisingly be an pretty strong partnership, because I think that compliment each other. Now granted, development on those two characters make it less and less likely that there can be something between the two and it's left to the Equestria Girls scene (Fluttershy and Spike the Dog is so insanely cute, I know that Spike is Twilight's dog, but I could really see Fluttershy as his owner if Twilight Sparkle was not in the picture).
Also, it would be a heck of a story where Pinkie and Spike got to be so close that they give each other's virginities--I really like the speculation that even though Pinkie is all bubbly and seductive and all into everyone grill, underneath she's a delicate girl, and it has to take a special friend for Pinkie to lead to her boudoir and do something so adult like lovemaking...

Good chapter. There were some parts that could be evaluated with the sense of scenery and such, like the elevator bit. You put a large paragrpagh for it and I thought more was going to come out of it, like a conversation on how he became the savior for the Crystal Empire or How did he become Twilight's Assistant. Stuff like that.
