Basically Pinkie was playing a game called "What" where you try to get someone unsuspecting to say the word "What" and when they do, you tell them they lost the game.

But Twi already said what when Pinkie burst in at the beginning:

“What is it, Pinkie?” said the purple egghead, listening intently. “Are we under attack again?”

So why didn't Pinkie just tell her in the first place?
