Let's see Luna is already teased and you wouldn't have genderbent Discord into Eris if you weren't considering it. The Mane 5 are obvious. Then the Canterlot 5 Moondancer, Minuette, Lemon Hearts, Twinkleshine and Lyra Heartstrings. Of course people would revolt if you broke up Lyra and BonBon/Sweetie Drops (threesome?) so might as well bring in the rest of the Background 6 (You genderbent Shining Armor why not the Doctor/Time Turner?). Everyone loves Trixie. Sunset Shimmer is a popular ship too. Then there's the challenge of Zecora and coming up with her dirty talk that just isn't rhyming with the word "cock," fifty times. That's 20 stories already I hope you have a lot of time on your hands.
You'll notice I didn't mention Starlight Glimmer. The main issue with her is that Twilight/Dusk is basically her parole officer if they have the same relationship they do in the show. That's an ethical minefield that would be very difficult to walk.
