Great story and i'd love it if you'd write a sequel to this story, but hey why not go deeper, sure have her return to him just not break-in. Have it that he realises something is off when walking through the everfree that Timberwolves bow and show reverance to him, or that somehow he can sense them nearby but notice them acting like guards for him in someway. Then during a full moon he has an eerie need to find his mate but ignores it for a while until his need to find her overtakes him and he goes slightly feral until one of the Timberwolves that are sent to guard/aid him break-in and take him to see his mate before he becomes a danger to himself and to those around him both pony and Timberwolf. So then he gets brought to her and nature takes its course, if you know what i mean. Then after their encounter she tells him that he should never ignore when he feels the need to be with her and always answer it when he feels it or he will become a danger to himself and others and that if he ignores this call to long he will go feral completely and lose himself to his Timberwolf instincts. Heck she could say that happened before and refer to the last Timberwolf that "prepped" him to mate with her and say he was another pony she mated with but after they finished he never answered the call in him for so long he eventually went full feral and transformed in that hybrid wolf that "prepped" him
I came here for porn, but I must admit I do have a scientific fascination with this method of reproduction. Technically, the timberwolf didn't ejaculate "sperm" cells so much as "specialized retro virus" cells. You know what's weird? That actually makes sense from wild survival stand point: why spend energy reproducing yourself when you can force other creatures to do it for you? Being able to redirect that energy into other functions could certainly help an organism survive, at least in theory. In fact, I now can't help but wonder if there are animals that do that in real life, perhaps in some obscure habitat. (There is still much we don't know about organisms in the oceans, so who knows?)
Then again, this would mean that an organism's survival depends entirely on the existence of other animals that happen to have compatible sperm and/or DNA: if a timberwolf pack can't get access to such a creature, than without a biological "Plan B", that entire pack is out of luck, which could effectively be an evolutionary trap, which - in the worse case scenario - could eventually lead to extinction.

First, the magnitude of the symptoms the Dragonpony suffers while under the Timberwolf's Curse (TC) can be explained by not been intended at all.
Dragons can increase their mass under certain stress situations, not unlike Hulk, and so can Tinker. It has to come from somewhere. The TC has an unexpected amount of nutrients to carry on its purpose. With any other host, TC would have to be very mild in order not to risk it's host's life. Tinker, though? It's time to go WILD!
Timberwolves in this setting act a lot like Parasites. At least one part of it's life cycle depends on a host. In this case, insemination.
The process reminds me a lot to Genestealers from Warhammer 40K, as it creates a hybrid offspring closer to the parasitic parent with some traits of the host.
Time to doing in the wizard!
Symptom: Arousal.
Purpose: Reduces aggressiveness of potential host.
Hypothesis of how: Phytohormones (Molecules created by a plant). The Host, if sentient, will interpretate them as the closest smell that induces arousal on it's species.
Symptom: Glowing sexual fluids/genitalia.
Purpose: A visual indication of TC activity. Very common in real world to signal expression of a product.
Hypothesis of how: A Scorpion primer inside monomers. A fluorophore located inside the RNA emits the light when active. Once the TC goes inactive, the RNA will fold itself and bring closer a Quencher that nullifies the fluorophore. Symptom: Elasticity of muscular tissues.

Capillaries will break, but that's what vascular growth factors are for. Second phase includes enhanced mythosis and growth of tissues. This allows for a more permanent stretching. Can also explain the abnormal growth of Tinker's genitalia too.
Symptom: Hijacking of seminal production.
Purpose: Primary purpose of TC, to continue the life cycle of the Timberwolves. Timberwolves are incapable of creating spermatozoa and needs the host to create the carrier cell instead. It allows to some of the host traits to be inserted into the timberwolf population.
Hypothesis of how: Lentivirus come to mind. They infect cells at all stages and insert the genetic payload inside the host genome. The stem cells are targeted. The glow of the modified spermatozoon indicates it's been infected with the TC.
Symptom: Swelling of testicles.
Purpose: Increased storage of fluids. Prostate is no longer used as it reduces bladder capacity, and urine destroys sperm, the main purpose of the process to begin with. The volume needs to be big enough to minimize the effects of the hosts immune system attempting to destroy the production.
Hypothesis of how: The testicle walls are slowly changed into tissue resembling those of the Bladder through reversing the differentiation and then re-differentiate into the several layers that made it. This can be done with some chemicals like Sox2 or Oct4. A bladder without space limitations can hold up to 10,5 liters / 2.6 gallons of fluid (It happened). Two of them can thus hold over 5 gallons of seminal fluid, enough to meet the "gallons and gallons" description provided.
Anal mating and 69:
Allow for fluids to better reach the circulatory system of the host and alter their physiology to better fit the TC purposes, such as nourishment, reduction of immunity system response or repurposing the host resources into the genitals and production.
Painless insertion:
By causing paresthesia (maybe because of the pressure of the growth), the host gets reduced nerve input and is left with just the pleasure of the stimulation and a tingling sensation. This reduces the aggressiveness of the insertion and makes it more compliant to further sessions of insertion.
