Ok, so, the general breakdown for anyone who doesn’t know the premise:

Unless I’ve missed it entirely (quite possible), there has been no “official” statement that Trixie is trans (and good luck getting HASBRO to admit anything close to this). But, while never explicitly stated, there are several hints that have made a strong case, and makes for completely valid head cannon.

Anecdotal evidence revolves around when Faust said there was an unspecified trans character in season one. There was also the revelation that Trixie was originally supposed to be portrayed as a stallion.

Next piece of evidence: Trixie is the only mare portrayed with the “stallion eye” effect.

But what probably drives the point home more than anything else, is that her color palette is that of the trans flag: Light Blue Coat, Pink Eyes (sometimes borderline violet, but there’s always pink in there), and White Mane.
