I think they're amphibious creatures with an interesting mixture of toad and Alligator traits.
4423962 Tarb would be a better person to ask than me, given he knows quite a bit about biology and stuff. My guess is that they are some form of Megafauna in the same category as Gojira (2014) and the Mutos.

But anyway, i think they descend from the same family as the Alligators given the structure of the adult's snout and that while capable of moving terrestrially, they prefer a more aquatic lifestyle and travel through the island by an underwater cavern system beneath the island with their main hub somewhere near the volcano(most likely dormant).
4423990 Given they have only three limbs in the form of two legs and a long tail, being amphibious would make sense. If they are reptilian then they must have a very long life span to grow so ridiculously huge especially if there is one that is the same size as Kong in the climax.

Crocs like the infamous Gustave are capable of exceeding 20 feet in length within the span of decades.
BTW Tarb, if you ever need more rifftrax for the preshow, TeamFourStar did one for Dragonball: Evolution. The only thing is, you'll have to manually sync the audio, the rifftrack is an audio file download, you don't actually get the movie with it (though honestly, who would want that movie).
