For me, comedy only suspends so much disbelief, so I'd say it only makes sense to me that Twilight would be honestly and absolutely furious. The only real change the comedy should make is how she acts on that fury.
Whereas in a serious story she would likely beat the living daylights out of her wayward friends, in this, she's likely to do something else instead.

She's come to love and cherish her friends, so that's going to go a long way to keep her from transforming into some psychopath just because she lost her pants. I highly doubt beating the daylights out of her friends is very likely in a serious story, or a comedy. I just don't see it in her.

Somehow, I doubt Twilight would care all that much for the game. In fact, the simplest solution present itself - make a quick series of teleport jumps to her own home, and in the morning, inform Pinkie and Co that she does not think she can be friends with them anymore.
