Orphan is definitely up there for one of the more disturbing bosses.
But Ludwig was the one boss that literally made me want to die when I got thrown into the fight lmao. Like, as soon as the cutscene is over, you realize just how fucked up he looks like and how big he is.
But, orphan is definitely disturbing as well. Definitely more disturbing on an emotional level too, considering the story.

If you mean the flood from halo, I think the ones you see in game aren't nearly as bad as several of the enemies in Bloodborne. As for the books and comics? Never read them.

Both are special for different reasons. The flood is a more natural horror, acting on similar principles to body horror movies and parasitic fungi. The creatures spawned by them carry an ever greater terror in the sheer degree of infectiousness they possess. They're literally an End of the World scenario.
The monsters from Bloodborne are different, in that they mostly attack the person mentally. They utilize special imagery and cosmic effects to wreak havoc on the fear center of the brain and warp your perception. The creatures spawned don't usually have a fungal look, and will instead have the flesh itself warp into vile abominations ranging from an angry fursona to walking flaps of spicy meat.
In general, the flood possesses the horror of a nearly unbeatable doomsday scenario, while the Bloodborne monsters utilize both the fear of the unknown and primal, cosmic terror. Both are almost equal if you can study the implications of each one, but by that same token we can see Bloodborne monsters come out ahead for this simple fact:
The Great Ones in Bloodborne can take over and empower the Flood.
So has Luna gone and told them all that he was never actually hostile, and all this trouble started because he was dropped into Luna's room by someone or something else? And that Luna is the one who attacked him first? Because that seems like pertinent information, that there was only danger because ponies tried to hurt him.

B. Celestia's being a bitch and dumping her problem on Twilight as usual (hilarious writing).
