Nightmare Moon has them trapped inside a dream, which would remove a lot of the need for physical power. The four stars aligning to let Nightmare Moon was a thing in the series premiere, so maybe she is using the alignment of the four stars to put ponies into a nightmare instead. In which case this could take place many years later, as in a dream you can alter Tot Te Hoi's size to when he was a 12-year-old, for instance
As for the potential first victim...I assume it'll be somepony who has a reasonably useful talent in solving the mystery of the school. So likely one of Blue Ray, who can seek stuff, A.K. Yearling, who probably has insight into tropes and cliches to apply to real life, and Caesar, who as a mathematician would also be a good logician. Secondary would be one of the two inventors.
