This is a surprisingly hard question for me. Not the Mane Six part of the question, of course ... I mean, PINKIE PIE, duh. Best pony foreveeeeer.

But I like so many of the background ponies! Hmm. Colgate's moved up my list ever since the I first started reading the "Ask Doctor Colgate" tumblr. Derpy actually got LINES, which is pretty rad. (Heh, lookit me, I'm still in the '80's ... gonna start saying "tubular" next ... ) Octavia's always good times, and Lyra and Bon Bon are adorbs. Not a big Doctor Who fan, so Doctor Whooves is kinda "meh" to me.

But y'know, even though she's gotten probably the least screen time of all the background ponies, her fanon personality more than makes up for it ... Vinyl Scratch, a.k.a., DJ P0N-3. She's my fave background pony.

I actually have it as a tossup between Twilight and Rarity for the mane 6... Twilight isn't as much of a complete bitch as Rarity tends to be, but I feel like Rarity can be a little less predictable and more fun most of the time. As for the background ponies I particularly like Cheerilee the best, she's got the most personality and the most screen time, as well as a whole lot of room for a dirty little secret... c'mon, schoolteacher!
