Difference between Gold and Starlight is that Starlight is acting out of concern for her friends. Gold is hiding things, and Starlight wants to know if those things are dangerous. She more than any other pony in Ponyville -bar Gold, irony of ironies- knows that a smile does not necessarily have good intentions. And in this regard, Manchineel might have pushed the wrong buttons in threatening Starlight. Before poisoning Starlight, Gold was a strange earth pony with an unknown magic and a phobia towards unicorns. Now though, Gold is a strange earth pony with an unknown magic and a phobia towards unicorns, and a phasing assassin ready to kill anyone who digs to deeply into Gold. That ups the threat level of Gold rather considerably to an outsider like Starlight.

might be a bit of an exageration to call Manchineel an assassin here (technically, she is, but that is beside the point). The most she did was irritate Starlight and make it so her Grand Daughter had a bit of privacy. If she really wanted her dead, she would've let Starlight follow Gold into the forest.

From an external perspective, sure, but to Starlight? She's just been hounded by somepony who can phase through object and uses various poisons to antagonize her, while also being threatened with death if she dug to deeply. "Assasssin bodyguard" would be a more than logical conclusion for Starlight to arrive to, and at the very least up Golden's danger level from "curiosity" to "potentially deadly threat."
