5345659 I've never heard of such a thing, but I think that with too much happiness, your body tends to speed up in certain parts and the rest of you can't keep up or handle it and ends up overloading you, thus killing you. That's my thought anyway and theory.

The Castle of the Royal Pony Sisters is the name of the old castle in the Everfree. Typically a castle and the town around it share a name, in this case Canterlot Castle is located in Canterlot. Conversely as the home of royalty and the seat of government it could just be called the Palace. However having the current residence of the only princess for a thousand years called 'The Castle of the Royal Pony Sisters' would raise a lot of questions and make Twilight a complete idiot for being surprised when Celestia called Luna her sister.

Twilight just said all that out loud. While it is possible she was thinking out loud and thus vocalizing her thoughts... that is not what the rest of the surrounding text implies. If in fact those are meant to be internal thoughts they should be in italics not quotation marks.

Overdoses can create situations where death is possible, most likely through affecting the blood pressure. But even if not fatal, gangrene of the hands and feet can also happen. With Equestria involved, who is to say that the brain's natural 'that's enough' switch couldn't break and flood the body with theses (in normal doses) useful chemicals. Also the process of death is likely to be horrible, with rapid unconsciousness being your best hope for relief.
