This EG/Human Granny Smith must be just like the pony Granny Smith in Tired Old Man's Old-Fashioned Bar Hoppers, elderly in age but still a rowdy teenager at heart. (Though IIRC she tries not to let her grand-kids know what kind of trouble she gets up to. Also IIRC "tries" is the key word there.)

At this point I believe she is both. She hatched his egg, raised him, tease him and confront Spike like sister and a mother would do. The only reason I don't think Hasbro don't have Spike call her mama is the same reason Bonbon and Lyra are 'friends' instead of a couple (and yes watching the equestrian game trailer seen below convince me of this). Worried parents will complain it sends the wrong image that they want little girls to be influenced too. Implied for older audiences to see, but not for younger audiences.

I mean I think it would be funnier if Cadanece is the one that tries to set up while Twilight is under the impression she is too young to date. It just be a great sense of humor with her arguing against it while Cadiance tries to set Twilight up.
