Seems that Sky's a bit pissed off with the world in general. I wonder if she got given Derpy as an employee, can't fire her, can't replace her with a more capable pony, and is horked off that the situation's preventing her from achieving the results she wants. Particularly if she's looking for promotion or a side-transfer to a more prestigious post office. She might also be horrified that her inability to deal with Derpy will reflect badly on her if she does attempt a career move - even if Derpy quits or Sky is promoted via outside intervention, that's not Skydancer dealing with the situation in a career-positive fashion, that's her struggling until external events conspired to free her.
To her, Derpy's sheer existence could be a constant reminder that her career aspirations may have been permanently trashed. That might cause a little stress in the workplace. I wonder if Sky's having problems sleeping, or she can be found in the Ponyville bars every night, or if she neatly, tidily, and efficiently goes home in the evening and spends an hour screaming into a pillow.
(Or maybe she only wants to run the Ponyville post office, but effectively, and Derpy is the barrier to that - one which could be lifted if Derpy quit and could therefore be replaced.)
It'll be interesting to see if 'Dancer is fleshed out into a 3D character, or if her role will be more Asshole Villain #6: Angry Boss Type.
