If I had to take a guess at what's going on... Queen Tiran was murdered by Chrysalis. However, she didn't stay dead as it were. The next fifty or so years was a spiritual battle for Chrysalis' body. If I had to guess, Chrysalis actually won the battle and forced the former Queen's soul out, trapping it inside of a gem... but the conflict forever altered Chrysalis and drove her utterly insane.

If I had to guess, the reason that Queen Tiran wants someone to get the gem to Canterlot is so that she can reclaim Chrysalis' body for herself and making a return.
