Mostly because the difference would be that if he did, he probably would have blown the planet up in a hissy fit instead.

Not knowing what OnePunch Man is, I really thought that Spike was just acting really well, and that his scales are particularly magic-resistant. I honestly thought he had scared Tirek by acting confident in his own abilities.

I think it might be due to being hatched by magical means and then being hit with immensely powerful magic turning him into an adult-sized dragon gave him some crazy resistances to everything. I'm planning on using it in my story to make him very difficult to hurt and make him much more like a Dragon type.

Interesting. If I may I'd like to speculate a bit... it's fun.
First, I don't think we've ever seen a time when magical spells have ever been used against Spike except when he was willing to allow them to affect him. (Dressing Spike up in a top hat and mustache comes to mind, as does Rarity's blinging him out in "Inspiration Manifestation.") Desire to be effected seems to play out in much of this. Spells that capture the girls also can capture him because he doesn't want to be separated from them.
Second, in that same, previously mentioned episode he digested.... literally digested a magical tome of immense power that was also tremendously corrupting. He hasn't suffered any known ill effects from that as yet. He could probably swallow the Alicorn Amulet and feel no pain. As he eats gems, and they are often a repository for magic, any magic he swallows is absorbed by the crystalline nature of his blood.
Third, I don't recall any time we've seen him in bandages. Dragon hide is obviously quite tough. Fire doesn't harm him and I'm not sure other elements would have much of an impact on him. What about Trixie turning him into a basketball in "Magic Duel?" Well, he wasn't physically altered so much as compressed into a spherical shape. I think that hint's at the malleability of dragon scales and hide, adding to his basic indestructibility. She obviously trapped him within a small, transparent sphere. Why would he allow it? Probably because he was still learning what could and couldn't effect him.
Fourth, we've seen what a fully mature Spike looks like in "Secret of My Excess." Noticeably, he's more wingless drake than Dragon and could probably give Godzilla a run for his money. In this episode he also showed he was resistant to pony magic.
Putting this all together, I would speculate that Spike is magic resistant at will. It's a power he slowly came to understand, probably by reading a lot of Twilight's books. When he desires, he can easily bat aside any destructive energy, be it magic or something else. More than this, he probably has the ability to act as a capacitor. Anything thrown at him can be countered and hurled back with either the same, or amplified strength. This is why he doesn't use much magic himself, as he mentioned in the story, but he is fully able to stand up to Tirek, whether attacked magically or physically. (If the bad guy stomps on him, the kinetic energy is absorbed then reflected right back at the attacker.) This gives him an awesome power and doesn't require much use of magic on his part. It also supports the One-punch man concept if he did actually store and release absorbed energy as desired.
Obviously if you are a villain, your best course of action when confronting Spike the Dragon is to ignore him. Of course, ignoring him would, over time, become harder and harder to do as he matures into a full sized drake.
Makes me wonder who the equivalent of Genos would be...

I admit until the end i was unsure whether you were going for one-punch man or for full powered ichigo from bleach, as Aizen accused ichigo of turning all his spiritual pressure into pure physical strength.
Looking back on it, both both shows have almost the exact same scene actually.
