Twilight Sparkle was one of the smartest kids at Canterlot High, always getting straight A's in all of her Classes. However, Twilight had to put her grades aside when the bombs started dropping as she and her friends were heading to safety. However, Twilight got separated from her friends in a sandstorm. When the storm cleared, she saw that she was in a crowd of people heading towards Vault 66. As soon as she put her Vault 66 Jumpsuit on, things were rather pleasant for her, as she had a room all to herself so she could perform further research and build more inventions.
However, she didn't know about Vault 66's true purpose, as it was an experiment by Vault-Tech to see how the human body would react to nuclear radiation. The result was that every dweller in the Vault was turned into either Ghouls or Super-Mutants.
Sci-Twi, out of fear, barricaded herself inside her room, but not only did the isolation and fear drive Twilight to insanity but some radiation that seeped into her room slowly but surely turned her into a Ghoul. Even though her body was flowing with radiation, her intelligence remained, and she joined the other ghouls and mutants in the new Mutate filled city of Feractopolis, which was the vault converted into a hotel/casino resort, where any mutant could come and relax from the harshness of the Wasteland. Sci-Twi works as the Bartender in the Bar/Casino side of the area. She even befriended a Giant Cazador and named it Spike.
Sci-Twi, along with the other Ghouls and Mutants inside, do not take kindly to raiders, or non-ghouls, and will capture and interrogate them on sight.
She also tells Sunset of the only thing that might be able to restore life and terraform the land, A Device known as "The Garden of Eden Creation Kit, or G.E.C.K. for short.
The only con to being a Ghoul is her decayed appearance, as it scares raiders away, but her flesh isn't really rotten. One pro to being a Ghoul is being immune to radiation, so highly radiated areas of water are like being in a heated swimming pool, with no injury at all.

"Oh don't worry Sunset, this pool of radiation is like being in a warm bath to me, it's not deadly to me at all."
