VERY definitely yes. As well as the rest of the Mane Six. After all, if Twi knew Spike and the rest of the Mane Six for at least a few years prior to Nightmare Moon's return, it would provide all the more reason for Twilight to trust them when the time was right AND all the more reason for Spike to consider Twilight a "geeky, but cool aunt". Visiting Ponyville twice a year (once during Celestia's visits during "Zap Apple Day" and once during Spike's hatchdays) on semi-annual field trips until Nightmare Moon's return would give Twilight plenty of time to gradually get to know all of them.
