I also want to know whence this comes. It looks like a an animatic from DHX, but it need not be genuine. Even if authentic, it could be a mistake which will be corrected during animation or it could be a joke which will not carry into animation.

It's from the clip from season five! And yeah, I agree it could well be an animation error (although...this is drawn, not animated :-/ )

After watching the movie, I notice that although the Mane 6 react to the CutieMarks, but not to the Alicorn. This supports an animation-error or joke which will not carry though to the final animation:
The show has 2 Primordial immortal Alicorns who are Goddesses, the 2 Royal PonySisters Princesses Luna and Celestia.Every know and then, ponies who achieve enlightenment go through apotheosis and ascend to Alicornhood. This happened to the PegasusMare Mi Amore Cadenza and to the UnicornMare Twilight Sparkle. These Alicorns are vastly more powerful and long-lived than regular ponies, but are still mortal and vastly weaker than the 2 Royal PonySisters.
It seems safe to say that the Mane 6 would comment in dialogue about an Alicorn, considering that less than 1 in a million ponies is even a mortal Alicorn and only the 2 Royal PonySisters are Immortal Primordial Alicorn-Goddesses.
