It usually takes at least a couple of hours, at the longest I've heard it take around three days. On average I'd say a day or so. It's a bit of a wait, and if your story has anything in it that makes it hard to approve (say, if it's tagged as a teen rated story and kinda pushes the line between T and M rating) then it could take longer because the proofreader might want to send it to another proofreader for a second opinion. If there's nothing complicated like that though, just a clearly rated story that's easily readable and doesn't have anything majorly controversial in it, then it'll be faster.
The good news is that once you have one story on the site, future stories you make won't have to go through the approval process, they just enter a short auto queue that takes around ten minutes.
