One morning, Twilight Sparkle discovers a strange package left at her doorstep. Curious, she sought out the contents (d'uh, wouldn't you?) and discovered a brand new game to share with all her friends, "Cardfight!! Vanguard".

If you're new to the game, I suggest you find an online manual to the game before trying to read...there isn't a detailed How-to-Play in the story, so yeeeah...

This looks promising, but could do with a little punctuation clean-up here and there.
My thoughts for when each pony becomes a card-fighter...
Twilight should pick up Royal Paladin. Why not, it's emphasis on gathering strengths from allies and has good consistency when it comes to calling from the deck, which suits Twilight's nature more than chance-based clans.
Applejack's independent streak would make her more suited for a Vanguard-focused strategy, so I always saw her as a Dimension Police user. It's still in a way connected to friends, as the Rear Guards are the ones to power up the Vanguard.
Rainbow Dash is a Nova Grappler user. Spam attacks over and over. Suits her.
The more erratic Pinkie Pie would relish (but not necessarily be good with) Pale Moon. Constantly appearing and disappearing is really her thing.
Rarity struck me as the type who'd want to have good control over the game and relish in a calculated battle of wits. That's why she'd use Oracle Think Tank.
Fluttershy wouldn't really use a fiercely aggressive strategy, so something defensive like Angel Feather would be something she'd enjoy. Or she could surprise everypony and use Spike Brothers (lol NO)

Everything you listed there is...absolutely perfect...my pals have been debating over this for quite some time. I agree with almost everything you listed there. I'm not sure, I've always pictured Twilight as the OTT-type, Rarity? I dunno really...Hmm, I'll have to discuss this again with my group. Thanks for your insight though, I'll be sure to implement your ideas.

I pictured Rarity as the OTT user since I figured she would employ a bit more method and care in her games. (Partially inspired by her disgust at Gears of War in the fanfic The Elements of Gaming, yet enjoying Command and Conquer.)
I also think it would be a good way to put her at odds with someone like Rainbow Dash, since rush tactics are OTT's biggest bane, so some of the story could be dedicated to the difference in fighting style. It could also be a good opportunity for Rainbow to learn to employ her tactics with more strategy. (I'm thinking about the long term effects of constant defense against something like Stern Blaukruger, for example).
Twilight needs to be the Royals user. Friendship is what the deck's about, and it's got a good amount of strategy for Twilight to work with as is anyway, since most of the calls from the deck are easily controlled, unlike the similarly themed Gold Paladin. Akane, Garmore, Alfred...you get the picture.

I'm still having doubts though, because the average CardFighter would immediately label Twilight as the OTT pony. I mean, just look at that picture! She's using Tsukuyomi for Celestia's sake. I'm afraid I'm going to have to go with Twi using an OTT deck because of "obvious reasons" and also to keep in sync with the picture, however, I'm still not sure on what to give Rarity. Perhaps an OTT deck as well, but with a different build? Maybe a Sakuya or a CoCo build? I'm still not sure, and until I figure this out, I won't be publishing any chapters of this any time soon.

Though I like the Spike of a MTG Story more, since he is playing a lot more cockier... But in that story were Twilight and Spike already playing for a while and not just starting...

Though please customize the decks later on... Oracle has a lot of possibilities for it.
Kagero is fitting for Spike. Though Narukami would have been a good choice too.

Also, I think that dedicating the first 3+ chapters just for an introductory Cardfight seems a bit much; the fight could be divided just as easily into simply two chapters. I mean you've only gotten as far as Turn 2...

At least you have constructive criticism, anyway; the reason for the short chapters is because this story is a mere experiment. I'm not entirely certain with my skills in writing a Cardfight/MLP Crossover and such... But, yeah, I suppose lengthening them won't hurt.
Also, the momennt you mentioned how I misspelled Bhar I facepalmed so hard there's an indent in my face. HOW COULD I MISSPELL MY FACE G1 VANILLA?!
