Use quotations when somepony is speaking:
"HEY WHO ARE YOU" screamed Twilight.
Use a type of punctuation inside the quotations:
"HEY WHO ARE YOU?" Screamed Twilight.
"HEY WHO ARE YOU," screamed Twilight.
"HEY WHO ARE YOU?!" Screamed Twilight.
Try a pause inside the speech, this seems like a lot to scream in one breath:
"HEY, WHO ARE YOU[insert chosen punctuation here]" S/screamed Twilight.
Just some pointers. Also, start new paragraphs whenever someone else speaks.
"What is it?" Asked Rainbow Dash.
"I don’t know," she said. "I think there is something wrong."
"Wrong with what?" Fluttershy said.
"I don’t know." Said Pinkie.
After a period ( . ) , question mark ( ? ) or exclaimation point ( ! ), the next word is capitalized. If it is a comma ( , ) then the next word, unless it is a name, remains lowercase.
Example:
"I don't know," said Pinkie. [/] CORRECT
"I don't know." Said Pinkie. [/] CORRECT
"I don't know," Said Pinkie. [X] WRONG
"I don't know." said Pinkie. [X] WRONG
