My only concern with the more obscure background ponies is coming up with a scenario to work off of, as many of them don't have enough of a character to really dedicate an event to them. If I'm unable to come up with a chapter idea that I'm happy with, they will probably be relegated to just a passing mention in some of the later chapters.

I assume you're going with the headcanon that Amethyst Star is also Derpy's daughter and thus Dinky's older sister (since they're seen having a picnic together in one episode and competing in the Sisterhooves Social together in another). Her I'd have no problem seeing getting knocked up.
Besides, according to one of the few bits of characterization she got in canon, she's a good organizer, so she and Twilight would probably bond over that.

The short answer is that Twilight's ascension is her basically going through a second puberty, but her libido specifically is dialed up like crazy because of her unique biology and her already pent up sexual needs. Even Celestia isn't aware of just how bad it is (don't worry, she'll find out). This will be addressed in story now that Twilight has had an outlet for her lust.

Maybe in a future chapter Twilight could plow the flower trio? She could either do them one at a time or all at once. If being an alicorn intensifies one's sex drive, I wonder what would happen if Twi met somepony who's been an alicorn longer than her like the other princesses. Would being an alicorn longer kinda mellow out their sex drives, or would they get more intense the older they get? I feel the Royal Sisters would be the dominant ones, and not let Twi have her way.

The princesses are going to be much more of a push-and-pull kind of scenario. Celestia and Luna's age and experience give them far more control over their own sex drives, but part of Twilight's excessive drive is the fact that she's a futa/hermaphrodite (I still haven't decided exactly which). Celestia is going to be unprepared for Twilight, that much is for sure. Luna already knows since she's seen Twilight's dreams, but is leaving her be until she feels the time is right.

Celestia is definitely a character that I've had several thoughts on how to handle her in this story. On one hand, Twilight's idolization of her for most of the series does make it hard to believe that she would be willing to try and dom her in the bedroom, but on the other hand I want to be careful about what characters can get one over on Twilight when she needs to be able to assert herself over characters like Chrysalis or Starlight Glimmer.
And yes, this story isn't going to be very favorable towards Starlight, I feel like she got off too easy when she was first redeemed and pairing her with a Trixie who absolutely got more crap than she deserved was not a good choice in my eyes, especially when Starlight would treat her extremely poorly
Personally, I think the CMC should all be on the same day, but not the same chapter:session. Also… maybe have the scent of her cock and cum be a trigger to change males to females? Would love for, after Twily finishes Rarity off, for Spike to start to turn shift to Barb by eating Rare’s creampie… maybe something happens and Big Mac changes in to Macarena <.< >.>

Just my opinion, but Luna's probably going to be delighted that the mare who's helped her out so much in the past is now finding happiness with many, many mares, and would gladly offer herself to Twilight to help her out. While being very amused by the currently youngest alicorn's turning the tables and domming the eldest.
By the way, any plans for Twilight to have a little fun with Cadance? (Not taking her away from Shining or breeding her, unless Cadance wants a second foal after Flurry Heart is born - just having a good time with her occasionally.)

I've been debating what I want to do as far as the male characters, as it kind of ties into something I'll be asking before one of the upcoming chapters. All I'll say for now is that if something can change them to female, it won't be permanent
