The time frame is too short, IMO. If Twilight only just 40, then so are her friends. It is well shown in MLP that the ponies must have near human lifespans. Being an Alicorn wouldn't mean she can't see or visit her friends anymore, especially since she's so lonely.
Now if you made Twilight 80-100 years old, it would make more sense since it would be reasonable to believe the rest of the mane six are dead, which is what you were hinting at already.

This. Granny Smith founded Ponyville, and Ponyville is over a hundred years old. The average lifespan of a pony IRL is 45 so I get where you're coming from, but show says different. I think maybe going way over the bounds of doubt with the age is best. Maybe 200-300 years.

In my head cannon, ponies only live till 50. I think this because their mentality is very childish and their culture and technology is shown to have advanced very little in 1000 years.
