Despite advice, I'm probably publishing a story with just the first chapter done.
But, a question: it's an epic detective story, so, "Comedy Drama Mystery," or "Comedy Drama Adventure"?

Drama, Mystery, Adventure sounds like the combo that best fits the description "epic detective story".
The descriptor "epic" along with the picture here make me think adventure is more fitting than mystery? I don't know that I can say much without having read the story, though, so whichever you feel is more appropriate for the first chapter is probably best.

I don't think I can get away with not labeling this Comedy, though. Think something akin to Pinkie perspective.

Picture is unrelated, but the story is essentially a detective story that focuses on a single missing pony's case the entire time, but it will involve travel and adventure outside of the initial setting.

I think that if your tone is consistent in the story it will be understood as Pinkie being Pinkie, since epic and detective already seem to have decided at least two of the tags.
Comedy, mystery, drama!
Ignoring the question you actually asked, is that picture from the Manga? It seems like that style. I really need to read it; apparently it's far better than you would expect it to be and isn't afraid to be memeworthy on occasion.

Would Comedy Adventure Mystery work, then? Mystery implies a certain level of drama, I'd say, but is more specific than a Drama tag, since most any conflict could be called Drama.

It is from the manga, although I don't know if it's an official colored panel, since the version I had was in black and white. And yes, the manga is amazing.
Fuck it, what you got to lose? Still better than publishing all chapters at once, anyway.

I'm leaning toward this, if only because it doesn't begin as an adventure. Mayhaps I'll call it an "epic mystery".

That sounds great!
Comedy drama mystery
I guess it all depends on if your trying to do a Sherlock Holms type thing where the reader needs to figure things out, or if your trying to do a Mike Hammer, Philip Marlowe, style gumshoe.
Figure out the clues, .......Comedy Drama Mystery
Dark Alleyways and car chases, .......Comedy Drama Adventure.

“I mean, you just seem to walk in random directions most of the time. How do you ever get to where you're going?" "Oh, that's easy," said Candy, with a smile, ”I just make the place I end up, the place I wanted to be all along."
