Twilight’s little smirk at the end of the short in almost a direct response to Spike’s uncertain groans of strain, effort, and despair in the face of an eminent long day (2:37) cements their relationship as sister-brother. She saw him sweating from exertion and had no comment besides an eye-roll and a little giggle. As an older brother, I recognize that little look of amusement / mild schadenfreude being directed at a younger sibling as they did something silly that I knew they would have to face the consequences for on their own, but I’ve never given that look to a younger child I was supposed to care for. A mother-son relationship between them would have prompted a more defensive response on Spike’s behalf, and at the very least a jab at Rarity for taking advantage of Spike and his feelings. Honestly speaking of this topic. I am shocked I have never seen a single fanfic I can think of in which Twilight just completely let Rarity have it for using and taking adventiage of Spike.
I would argue that depends on what (psychological) age Twilight thinks of Spike as there. I have 100% seen mothers of teen boys get that look when their sons are, for example, doing more physical labor than they've done in their life to save money to take out a girl, when a week ago they had to be threatened with grounding to get them to pick up their shoes. It's clear she doesn't see Spike as a baby dragon from a mother's perspective (at least, not anymore) but I don't think it's out of line with seeing him as a teenager.
(I'm not married to one view or another, just I don't think your reasoning shuts down the opposing view.)

This is a good point. Everypony else thinks he’s pretty responsible. Watched Ail-icorn and everypony else was like ‘yep good luck Spike you got this’.
Also cute baby alicorn is now canon so that’s pretty cool.
