A forewarning though, there's already been quite a few "Spike is dating X and Twilight gets jealous" type of fics

Spike trying to avoid Pinkie Pie finding out that Twilight and he are in love could make for some great shenanigans if you're going for the comedy route.
Spike trying to break the news about his relationship with Twilight to Fluttershy could make for some heart-wrenching tears.
Spike trying to avoid being murdered by Rainbow Dash she she finds out he boinked Twilight would make for an excellent scene (This could go with either of the above two really.)
EDIT: Feck it, I'm going to play around and see which Spike shippings aren't done frequently.

Well... Pinkie is fine to write for, but i look at her as a difficult subject because no one can really write her down as random as she should be without overdoing or understating her personality.

I can't suggest anything about her, but when I was in Driver's ed, my teacher told us that if we are bad at reverse try to avoid situations where reversing is necessary all the time.
Spike being 16 gives more opportunities than just the mane 6, Sweetie Belle is a bit overused, Applejack and Rarity seem like they wouldn't care if Spike cheated on them, unless you did a backstory of how well their love was.
If you want to make it so the pony is sad for being left, Fluttershy is best option, if you want jealousy, Rainbow Dash.
So that leaves you with one option, Raindrops. Raindrops is always the best option.

Fluttershy is alright, but mystery endings are cool too. But if you go with Applejack, there might be some racism (him being a dragon and all) from the Apple family that might be fun to exploit. Plus, that can be Twilight's doomsday device she has to result to or has to confront.
I would suggest going with Pinky. I can see her as being well and truly devastated in a way that she just can't recover and have the rest try and recover as Spike moves on to be with Twilight. Nothing against poor Pinky but if your going for the hard mature rating her just breaking and wasting away would be a good route. I always imagined Fluttershy and Spike would be more master and pet in a overly cute way.
