Let this begin with an accidental drug trip from a botched experiment in her lab.
Make everypony write a full biography of themselves so twilight get a bunch of new books to read

Ah. Because website tends to be less trigger happy when you're zapping non-anthros in your imagination inspired by text on a screen.
I can see Twilight using hyperbole and figures out speech that she unwittingly warps reality with.
Like saying a pony who is working relentlessly is secretly a robot.
Or something she says in total sarcasm that becomes true.
"Applejack works so hard each day every day."
"Like she's secretly a golem just programmed to pretend it has a personality?"
"Oh yes, that is totally right."
"Do you PAY Spike for the work he does for you Twilight? You know what unpaid workers are called right?"
"Right, Spike is totally my mindless slave who I just let ponies think has free will."
Or the CMC's antics cause trouble again,
"Ugh! Those fillies aren't ponies! They're little imps who serve Discord!"
(With the imps realizing Twilight's power, and leading her on in her choice of words to warp reality MORE just for the fun of it! They're aware of who and what they were before, but don't give a damn.)
Or seeing Big Mac for one reason or another dressed up as Red Gala, or someone seeing a picture him as Red Gala and Twilight jokes, "Yeah, Red Gala is most definitely the most beautiful mare in Ponyville!" Cue Red Gala being real and Big Mac a disguise.
"Just you wait Twilight! Trixie WILL surpass you!"
"You'll be an Alicorn princess someday, Trixie, I'm so sure."
(Twilight says someday, so it doesn't happen at that spot, but fate is now set that Twilight will day undergo Apotheosis).

Quadruped is always going to be better than anthro, but generally speaking, they should always be nudists.

I would rather nice things happen to characters. Like she says to spike that if he eats his vegetables and gets his exercise he'll grow up to be big and strong, and then we see him later after he's eaten his vegetables and gotten his exercise and he's bigger and stronger. Or she says that lots of ponies will be wanting something like a dress that Rarity made for her, and then a bunch of ponies want something like it and she has business. Or she sarcastically says that Fluttershy and Discord are going to get married, and then we jump to like a few days later when she's begrudgingly sitting at the wedding of Fluttershy and Discord.
Could also have her say that a male character is a female, and we later see them and they're embarrassed that they're now female, or vice versa.
What I would actually really like to see here, because it makes sense, and is a road that people don't normally go down, is her suddenly realizing that something is up, figuring out that things she says are coming true, and then trying to do a bunch of experiments to see what sort of power she actually has here and trying to figure out what sort of magic is behind it, but also maybe having a bit of a crisis asking the ethical implications of just changing things in reality.

Scootaloo becomes Daring Do's younger sister who helps her out on adventures.
And a few other ideas:
Lightning Dust becomes Spitfire's maid
Zephyr Breeze's affections towards Dash get transferred to Lightning Dust, so he becomes Spitfire's butler so he can work alongside her.
Pinkie and Shy swap personalities, but keep their careers. So Shy becomes an outgoing animal lover, while Pinkie becomes a shy baker.
Later on, Dash gets turned into captain of the Wonderbolts with Fluttershy joining the team because of her, Spitfire becoming their third in comma
Got another few (i hope it's okay that I'm making so many in a row. I can't really control when inspiration strikes):
Rarity gets turned into Dash's maid for a while
Sweetie Belle and Apple Bloom end up thinking that AJ and Rarity are their sisters (meaning they swap)
Shy's parents become much more bold and brave.
The pets all become anthro versions of themselves.
Angel Bunny becomes much more polite.
Spike gets turned into Dash's assistant instead of Twi's, but his crush on Rarity remains

Chrysalis becomes a kindhearted maid for Shining and Cadance
Okay turns out my brain decided that there needed to be a few more ideas:
Gilda gets turned into a soft spoken ballerina
Garble gets turned into Spike's personal butler
Ember gets turned into a cheerleader

Yeah, see, that's the hard part.
I'm drawing a complete blank on that aspect.
Only one I can think of an explanation for is the idea I posted on the story itself, where Twilight, upon seeing how much more relaxed Dash is after a little bit as a fashionista, decides to experiment and see how being turned into a tomboymakes Rarity feel.
